研究興趣

1.  主要研究梭狀桿菌表面蛋白的致病性,特別是Clostridium difficile(困難梭狀桿菌芽孢桿菌)的研究。 C. difficile是一隻具有多重抗藥性的人類致病菌,也是引 起住院病人抗生素相關性腹瀉排名第一之致病原,過去的數十年間,由於高致病菌株的出現,發生率持續地增加。許多 文獻指出,外毒素(TcdA 與 TcdB)是造成抗生素相關性腹 瀉,甚至危及生命的偽膜性腸炎的最主要致病因子,雖然如此,其他與附著和繁殖有關的致病因子所知甚少,因此我們 嘗試著揭開這些與致病相關的表面蛋白分子之神秘面紗,試圖了解sortase-anchored蛋白在附著與繁殖上所扮演的角色,其次,我 們將會探索C. difficile Type IV Pilus (T4P) operon的結構與相關功能。

2.  與sortase有關的技術,sortase是一種轉肽酶,幾乎存在於所有的革蘭氏陽性細菌,sortase辨識具有LPXTG motif 蛋白的C’端,並將它固定於細胞壁新合成的肽聚醣上。大家所熟知的益生菌如Lactobacillus longum(龍根菌)及L. casei都具有同源的sortase,我們想利用基因重組工程將致病原蛋白表現於益生乳酸菌細胞壁表面來發展”喝的疫苗”。這種傳遞方式的便利性及疫苗的穩定性,使得宿主能夠建立對抗外來病原的抵抗力,以增強自身的免疫系統,深具發展的潛力。
