Abstract

Signal transducer and activator of transcription 3 (Stat3) is a transcription factor which plays a critical role in the regulation of cell survival,proliferation, differentiation, apoptosis and angiogenesis. Constitutive activation of STAT3 was detected in a wide range of cancer cell types and showed a poor prognosis. However, the specific negative regulator of STAT3 has not been found. In this paper, the authorsdemonstrated that the chaperone protein GdX mediated an interaction between phosphatase TC45 and STAT3 in the nucleus. This interaction initiated STAT3 dephosphorylation, leading to downregulation of STAT3 transcriptional activity. Moreover, they showed that overexpression of GdX suppressed tumor growth and metastasis. In summary, these data indicated that GdX functions as a tumor suppressor on STAT3 activity. This provided the value of the specificity of GdX on TC45-regulated STAT3 dephosphorylation as a new strategy for anti-cancer therapy.
