A Novel Role for PECAM-1 (CD31) in Regulating Haematopoietic Progenitor Cell Compartmentalization between the Peripheral Blood and Bone Marrow

Platelet endothelial cell adhesion molecule-1 (PECAM-1/CD31) is a molecule expressed very high levels on the surface of endothelial cells and at moderate levels on most hematopoietic cells. PECAM-1 acts as an important factor in regulating cell adhesion, migration and activation in haematopoietic cells, and contributes to several important processes in vasculature. In previous studies, the authors found PECAM-1 can control megakaryocyte migration by polarization of CXCR4 and regulation the adhesion to bone marrow matrix proteins. The haematopoitic stem cells (HSCs) mobilization or homing between born marrow and circulation has been shown to be regulated by CXCL12 (SDF-1) and its receptor CXCR4 interaction. In this paper, they wanted to investigate the physiological role of PECAM-1 in the bi-directional movement of HSCs between the vascular niche and circulation. The experiment results demonstrated PECAM-1−/− mice exhibited an increase in the number of circulating progenitor cells in the resting state due to defect of HSCs migration to CXCL12 and increased levels of MMP-9 in bone marrow. These findings further support a novel role for PECAM-1 in modulating cellular migration and adhesion and as a regulator of HSCs mobilization.
