Abstract:

Epithelial–mesenchymal transition (EMT) occurs during critical phases of embryonic development in many animal species, and it is essential for early stage of tumor metastasis (1). Some reports used different cancer cell lines to find out miRNAs related with EMT, but these can not represent EMT in normal situation. The authors used MDCK cell line to observe the EMT process, and found that microRNA-200 family (miR-200a, miR-200b, miR-200c, miR-141 and miR-429) and miR-205 were markedly downregulated in response to transforming growth factor (TGF)-β or to ectopic expression of the protein tyrosine phosphatase Pez (PTP- Pez) (2). These miRNAs can regulate ZEB1 and SIP1, E-cadherin transcriptional repressors, by 3’UTR reporter assay. The authors reversed EMT or mesenchymal to epithelial transition (MET) by changing the levels of miR-200 family and miR-205. They confirmed the expression of these miRNAs in breast cancer cells and breast cancer specimens, and proved miR-200 family was lost in mesenchymal cell lines and clinical samples. These data indicate that downregulation of these miRNAs is an important step in EMT and tumor metastasis.
